In vitro assessment of agave fructans (Agave salmiana) as prebiotics and immune system activators


Abstract
The prebiotic effect of agave fructans (Agave salmiana) was evaluated through the growth of two lactic acid bacterial (LAB) strains (Lactobacillus casei
and Bifidobacterium lactis).
The immune system was acti-vated through the stimulation of peripheral blood mononuclear cells (PBMC) of healthy subjects testing fructans, LAB or a mixture of these compounds at different concentrations.
Immune responses, such as early cell activation (CD69), cell cycle progression, nitric oxide (NO) production
and the expression of transcription factors for lymphocyte differentiation, were analyzed.
Compared with other fructans, the extracted agave fructans showed the highest prebiotic activity
and increased levels of CD69 expression,proliferative activity
and NO production when administered with the probiotic L. casei.

The Th1 lympho-cyte differentiation produced through LAB stimulation was greatly diminished after the incorporation of agave fructans.
In conclusion, these types of fructans (A. salmiana) are involved in the activation
and selective differentiation of cells of the immune system through interactions with probiotics.
Thus, agave fructans represent a novel immunomodulator that might benefit the functional food industry.


1. Introduction
Fructans, a heterogeneous mixture of fructose polymers linked by fructose–fructose glycosidic bonds [1], are increasingly being added to foodstuffs
and medical treatments with claims of health benefits [2,3].
In Mexico, fructans are isolated from the agaves, as the main photosynthetic product stored in the stem of these plants [4].
Due the structure
and type of link ( 2-1 and 2-6) of these molecules, fructans are considered as prebiotics [5].
Fruc-tans are not digestible through intestinal tract enzymes,
and whole molecules pass into the colon, where these polymers become fer-mented through beneficial intestinal microflora, also known as probiotics or lactic acid bacteria (LAB), favoring the growth of these microorganisms [6–9]. This activity induces many health benefits, such as reduced blood glucose levels
and increased lipid homeostasis, mineral availability
and modulation of immune func-tions [10–14].
Some studies on the immune system have reported the regulation of intestinal micbiota, increased resistance to infections,
and the reduced activity of gastrointestinal diseases,
allergic reactions and colon cancer cells [13,15–17].
Fructans have been associated with the immune system through the recognition of the molecular patterns of LAB in the intesti-nal microflora, which affects the composition of the gut-associated lymphoid tissue (GALT).
Intestinal epithelial cells detect peptido-glycan in Gram-positive bacteria through Toll-like receptor (TLR2)

and major histo-compatibility complex I and II molecules, result-ing in the activation of a complex intracellular signaling cascade,the up- or down-regulation of inflammatory genes
and the pro-duction of pro- or anti-inflammatory cytokines, initiating
and regulating both innate
and adaptive immune responses [13,18].
Bacterial signals can be transmitted to adjacent immune cells, such as macrophages, dendritic cells
and lymphocytes [15,18].
The main effect of probiotics is to increase the functions of innate immunity,including phagocytosis
and cellular cytotoxicity mediated through
natural killer cells (NK) [19,20],
and regulate the synthesis of cytokines inpolarized T-cells [21,22]. Due the regulatory T cellsplay a significant role in dampening misdirected immune responses,probiotics might exert anti-inflammatory effects through influenc-ing this T-cellpopulation[23].
T lymphocytes, Thelper lymphocytes CD4+ (Th)
and cytotoxic T lymphocytes CD8+ (CTL), are function-ally distinct cells depending on the type stimulation received from cytokines.
T helper cells can be subdivided into Th1, Th2, Th17
and regulatory T cells (Treg) through of the induction of spe-cific transcription factors, such as T-bet, GATA3, RORC
and FoxP3, respectively, to define an immune response [24]. Th1 cells medi-ate the cellular immune response through the production of the cytokines INF-
and TNF-· Th2 cells mediate the humoral immune response through the production of the cytokines IL-4 and IL-10.

Th17 cells are involved in the induction
and progression of sev-eral proinflammatory processes,
but the precise function of thesecells remains controversial.
Moreover, Treg cells regulate the activ-ity of other T cells upon completion of infectious or inflammatory processes [12,24].

The beneficial health effects attributed to the intake of prebi-otics have been reported in several in vivo
and in vitro studies [14,16,25–27].
However, little is known about the direct action of prebiotics with the immune system [15,28],
and particularly, there are no studies concerning the effects of agave fructans on immune cells.
Therefore, the aim of the present study was to evaluate the in vitro activity of agave fructans (Agave salmiana), as prebiotics
and immune system activators, on peripheral blood mononuclear cells (PBMC) from healthy subjects in the presence
and absence of LAB at different concentrations. The results of these studies using agave fructans were compared with those of other studies using the widely known
and distributed inulin-type fructans from chicory.


2. Materials and methods
2.1. Fructans and microorganisms
Extracted A. salmiana fructans, commercial chicory inulin-type fructans (Beneo Orafti® LGI, Tienen, Belgium)
and reagent chicory inulin-type fructans (Sigma–Aldrich, St. Louis, MO, USA)
were used, and the specific characteristics of these compounds are listed in Table 1 [4,29,30]. Two lyophilized strains of lac-tic acid bacteria (LAB), Bifidobacterium lactis (SACCO BLC1, RAFF,Zapopan, Jal, Mexico)
and Lactobacillus casei (SACCO BGP93,RAFF, Zapopan, Jal, Mexico), were used. Lyophilized L. casei was reactivated in 20 mL of Man-Rogosa-Sharpe broth (MRSB;Becton-Dickinson Difco, San Jose, CA, USA) using a inoculation loop
and incubated at 37 ◦C for 24 h, while lyophilized B. lactis required two sequential reactivations of 24 h in MRSB supplemented with 0.5 g/L l-cysteine (Analytica, Monterey, NL, Mexico) under the same conditions.
To characterize the growth phases of both strains, a 24 h growth curve for each microorganism was performed in a 1-L flask of MRSB,
and samples were collected at different time periods (0.5–1 h intervals).Microbial growth was determined through plate counting using the Miles and Misra method [31].
The growth curves for both bacteria showed similar behavior, reaching the station-ary phase between 12 and 15 h.
Therefore, 24 h incubation was enough to achieve stable bacterial growth in subsequent assays.
For the immunological stimulation of cells, both bacterial strains were grown to the stationary phase, washed
and resuspended at 1 × 1010 CFU/mL (Log 10 CFU/mL) in sterile phosphate-buffered saline (PBS).
To evaluate the concentration of LAB, six sequential dilutions at 10% of this bacterial suspension were performed in PBS to obtain inocula at different concentrations (Log 8, Log 6 and Log 4 CFU/mL),
and the inocula were stored at 4 ◦C during the study.
The concentration and bacterial viability of each inoculum was verified through weekly plate counting, which remained constant for 45 days.
2.2. Assessment of the prebiotic activity of fructans The prebiotic effect of fructans was evaluated trough the B.
lactis and L. casei growth.
The response as prebiotic was evalu-ated using a culture media with the same composition of MRSB

but replacing the carbohydrate source for the fructan type (agavefructans, commercial inulin or reagent grade inulin).
This bacterial growth was also evaluated incommercial MRSB and inthe prepared MRSB without carbohydrate source (MRSB-WCH). These assays were used as controls. The prepared MRSB contains 10 g protease peptone No.3 (Becton-Dickinson Difco, SanJose, CA, USA), 10 gmeat extract (Becton-Dickinson Bexon, DF, Mexico), 5 g yeast extract(Becton-Dickinson), 1 g Tween 80 (Química Meyer, DF, Mexico), 2 gammonium citrate (Química Meyer), 5 g sodium acetate (Analytica,Monterrey, NL, Mexico), 2 g dipotassium hydrogen phosphate 2.0(Sigma–Aldrich, St. Louis, MO, USA)
and 0.1 g magnesium sulfate(CTR, Monterrey, NL, Mexico), diluted in a liter of distillate water.
The fructans concentration in all the assays was 20 g/L. All the broth culture media for B. lactis were supplemented with 0.05 g/Ll-cysteine.
The activated LAB was inoculated with 0.6% (v/v), cor-responding to approximately 1 × 108 CFU/mL in different culture media (25 mL)
and incubated for 24 h at 37 ◦C.
The precise con-centration of each inoculum was verified through plate counting.

Samples were collected from each assay
and plated onto MRS agar using the Miles
and Misra method [31], and the bacteria were incu-bated anaerobically at 37 ◦C for 48 h. The growth ofthe bacteria was quantified through plate counting
and expressed as Log CFU/mL.

All assays were performed in triplicate yielding a total of 30 exper-iments.

2.3. In vitro immune stimulation of PBMC using two microorganisms
and two fructans 2.3.1.
Cell culture
Peripheral blood mononuclear cells (PBMC) were isolated using the Ficoll-Hypaque (Sigma–Aldrich) technique, washed
and cul-tured at a density of 1 × 106 cells/mL at 37 ◦C and 5% CO2 in 12-well

plates (Corning Costar, Corning, NY, USA) containing RPMI 1640 culture medium (GIBCO, Grand Island, NY, USA) supplemented with 10% fetal bovine serum (HyClone Laboratories, Logan, Utah,USA)
and 2 mol/L l-glutamine (Sigma–Aldrich) without antibi-otics.
The PBMC were stimulated with agave fructans (extracted from A. salmiana), chicory inulin-type fructans (Sigma–Aldrich), L.casei, B. lactis or different mixtures of fructans/LAB at the indicated concentrations according to the experimental design.
2.3.2. Early cell activation Early cell activation was measured through the expression of CD69 by flow cytometry analysis. After 24 h of incubation,cultivated PBMC were immunostained with mouse anti-human CD69-phycoeritrin (PE)-labeled monoclonal antibody (Becton-
Dickinson, Bioscience, San Jose, CA, USA), washed with PBS, fixed with 1% p-formaldehyde,
and analyzed using a FACSCalibur flow cytometer with Cell Quest software (Becton Dickinson).
The results are expressed as the percentage of positive cells.
PBMC stimulated with phorbol myristate acetate (PMA; 20 ng/mL; Sigma–Aldrich) were used as a positive control for this technique.

2.3.3. Cell proliferation or cell cycle

The cell cycle of the treated cells was determined accord-ing to the nuclear DNA content through flow cytometry.
After 72 h of incubation, cultivated PBMC were washed with PBS
and resuspended with vigorous stirring in a hypotonic fluoro-chrome solution (propidium iodide in sodium citrate, plus 0.1% Triton X-100).
The samples were incubated at 4 ◦C in the dark for 30 min prior to the flow cytometry analysis. The results were expressed as the percentage of cells in distinct phases of the cell cycle: G0/G1 (normal diploid DNA phase), S (DNA synthesis phase), and G2/M (mitosis phase).
PBMC stimulated with phytohemagglutinin (PHA;10 g/mL; Sigma–Aldrich) were used as a positive control for this technique [32].

2.3.4. Nitrite concentration
The nitric oxide (NO) production in cells was estimated through the measurement of nitrite (NO2−) using a microplate assay based on the Griess reaction. 100 L of supernatant aliquots of each assay were incubated with 10 L of Griess A reagent [0.1%, w/v N-(1-naphtyl)-ethylenediamine hydrochloride (NEDD)]
and 10 L of Griess B reagent (2%, w/v sulfanilamide in 5% HCl) at room tem-perature for 20 min. The absorbance at 540 nm was determined on a Synergy HT Multi-Mode Microplate Reader (Biotek, Winooski,VT, USA).
NO2− was determined through calibration curve using sodium nitrite as a standard at concentrations from 0 to 200 mM.
PBMC stimulated with lipopolysaccharide (LPS; 5 g/mL; E. coli;
Sigma–Aldrich) were used as a positive control for this technique[33].

2.3.5. Experimental design A distance-based experimental design was performed to obtain the principal effects of 2 categorical variables (LAB and fructans type), 2 continued variables [LAB concentration (CLAB)
and fructans concentration (Cfructans)],
and the first order interactions between them. Six blocks were selected to eliminate the unwanted effect of interindividual variability, yielding a total of 30 experiments as shown in Table 2. The response variables included the early activation of lymphocytes (percentage of CD69 positive cells), the proliferation in cell cycle (S or G2/M) and NO production.
2.4. Transcription factors expression in lymphocyte differentiation of stimulated PBMC
TotalRNA was isolatedfrom2millionPBMCusing TRIzol reagent

(Invitrogen, Life Technology, Carlsbad, CA, USA),
and the concen-trations
and integrity of the RNA sample were measured using a spectrophotometer Synergy HT (Biotek). cDNA was generated with 100 ng of total RNA using the High Capacity cDNA Reverse Transcription Kit (Applied Biosystems, Foster, CA, USA) accord-ing to the manufacturer’s instructions. A total of 100 ng of cDNA was applied to real-time PCR using predesigned TaqMan gene expression assays, according to the manufacturer’s instructions (Applied Biosystems).
The following probes were used: TBX21,Hs00203436/m1; GATA3, Hs00231122/m1; RORC, Hs01076112/ml;and FOXP3, Hs01085834/m1.
The mRNA levels were normalized to 18S as an endogenous control.
All reactions were analyzed using a 7900HT CFX96 Real-Time PCR System (BioRad, Hercules, CA, USA).
The cycle threshold (Ct) values, corresponding to the PCR cycle number at which the fluorescence emission reaches a threshold above baseline emission, were determined for the basal expression of mRNA.
The relative mRNA expression was calculated using the 2−Ct method for the mitogen-induced expression of mRNA in relation to unstimulated cells [34].
2.5. Statistical analysis
The statistical analysis was performed using the Design Expert 7 (Stat-Ease, Minneapolis, MN, USA) statistical package.
The results are reported as the means ± standard error (SE)for parametric data.
Analysis of variance (ANOVA)
and comparison of the means were conducted using Tukey’s test.
A probability value of p < 0.05 was considered significant.

3. Results and discussion
3.1. Assessment of the prebiotic activity of fructans The principal characteristic of prebiotics is the ability to enhance the proliferation of beneficial microbes or probiotics in human colonic microbiota [10]. In the present study, we analyzed the prebiotic activity of the extracted A. salmiana fructans (Fructan A.s) compared with that of conventional prebiotics preparations.

The results were expressed as growth of LAB (L. casei and B. lac-tis)
and are showed in Fig. 1. The results were calculated as the difference in CFU/mL between the strain growth
and the inoc-ulated quantity (CFU/mL); thus, the height of the bar indicates the net growth after 24 h, expressed in Log CFU/ml ranging from 8.1 to 9.3 Log CFU/mL. In all the fructans tested, enhanced bacte-rial growth (p < 0.05) with respect to the negative control (culture media with the same composition of commercial MRSB but with-out a carbohydrate source; MRSB-WCH) was observed. For L. casei,there were no statistically significant differences between either the tested fructans or the positive control, but all bacterial growth ranged from 9.07 to 9.2 Log CFU/mL compared with the standard commercial media MRSB, indicating high prebiotic activity.
How-ever, for B. lactis, the type of fructans might be an important factor (p < 0.05), as the agave fructans showed the highest bacte-rial growth (8.8 ± 0.4 Log CFU/mL)
and consequently the highest prebiotic activity.
Differences in bacterial growth due to the strain type were observed, where L. casei constantly showed higher growth than B.lactis (p < 0.05). These results were consistent with other reports confirming differences between L. casei and B. lactis growth using saccharides [35]. Differences in the ability of each strain to use dis-tinct prebiotic preparations might reflect the wide phylogenetic

differences between L. casei
and B. lactis, as these bacteria belong

to different phyla (Firmicutes and Actinobacteria, respectively) [18].
There was also an interaction between fructans type and the strain
used (p < 0.05), where all the fructans tested were more selective
to the type of strain, favoring L. casei. In addition, it was described
that prebiotics might selectively stimulate the growth and activity
of one or more bacterial numbers [10]. Therefore, agave fructans
might promote L. casei growth more than B. lactis.

Others in vivo studies have reported bacterial strain dependence in the presence of fructo-oligosaccharides [35].
In addition, it has been reported
that lower degree of polymerization (DP) fructans more efficiently support the growth of B. lactis than high DP substrates [36].
These data suggest that the agave fructans induce LAB growth same
or better than commercial fructans.
3.2. In vitro activation of PBMC using two microorganisms and two fructans
Because a fundamental immunomodulatory role of the pro-biotics
and prebiotics on cells and immune responses has been previously demonstrated [13,25], in this study was investigated whether agave fructans activate the immune system
and induce cell proliferation.
Table 3 shows the significant effect of differentfactors , such as CLAB (A), CFructans (B), LAB type (C), fructans type (D)
and the interactions between them, on each response vari-able.
The early activation of the lymphocyte population in response to stimuli (fructans and LAB) was measured as the percentage of CD69-positive cells.
Lymphocyte activation depends on CLAB (A),
the fructans type (D) tested, the interactions between the LAB strain
and fructans type (C × D) and their respective concentrations(A × B).
The C × D interaction is shown in Fig. 2a for different fruc-tans/LAB combinations.
The agave fructans/L. casei mixture (F + L) showed increased activation, with 10.8% of CD69-positive cells.

Fig. 2b shows the response to the A × B interaction for the F + L mix-ture, where the activation increased with higher CFructans
and lower CLAB, with 12.4% of CD69+ of lymphocytes.
These results are consis-tent with previous studies showing increased CD69 expression in CD8+NK
and CD8+T cells after stimulation with Lactobacillus strains [37].
In addition, other studies have shown a significant increase of CD4+CD25+ (T-helper activated regulatory cells), CD8+CD25+ (T-suppressor/cytotoxic activated cells),
and CD16+CD56+ (NK cells) upon stimulation with probiotics strains [38].
These data sug-gest that the F + L mixture shows immunomodulatory effects on immune cells.
However, we assessed the effect of fructans on the PBMC cell cycle.

Fig. 3a describes a typical DNA histogram, showing three cell populations (G0/G1, S and G2/M) based on the DNA content.
From these phases, only the S phase was significantly affected (p < 0.05)
in response to the CLAB (A), LAB type (C)
and the interactions A × B,B × C
and B × D, while with the type
and concentration of fruc-tans (B and D) showed no effects.
Nuclear DNA replication occurs in the S phase, indicating PBMC proliferation, which was induced predominantly through LAB in a dose–response manner. The cell
percentage in the S phase from PBMC assays stimulated with LAB
and fructans mixtures is represented in Fig. 3b, where agave fruc-tans/L. casei mixture (F + L) showed the highest percentage at 11.9%, which was greater than that shown for the positive control (PHA)(p > 0.05).
In vivo studies have described the increased proliferation
and activity of CD56+ NK cells in peripheral circulation after the consumption of LAB strains in elderly subjects [20],
and induced murine splenocyte proliferation was observed in rats after LAB con-sumption [39]. Furthermore, it was reported that Bifidobacterium longum was effective for induction of IgA+ cells proliferation in small
and large intestine of mice during infection [40].
The results obtained in the present study showed that the fructans/L. casei mixture induced the proliferation of immune cells.

Moreover, we measured nitric oxide (NO) synthesis to deter-mine whether agave fructans induced monocyte activity. The nitrite concentration in the supernatant was determined as an indi-rect measure of the synthesis of NO, a mediator of many immune
and inflammatory responses, produced exclusively from mono-cytes/macrophages.
Table 3 shows that the CLAB (A)
and type of LAB (C) are the principal factors that positively affect NO production.

Fig. 4 shows the C × D interaction, for nitrite concentration at 24
and 72 h of stimulated PBMC.
The greatest production was observed at 72 h of incubation in all cases,

and 18.3 М NO2 for 24 and 72 h respectively.
These results were greater than those obtained for the positive control (LPS) (p > 0.05). Reactive nitrogen intermediates might be involved in the regulation of monocyte function, showing immunoregu-latory effects relevant to the control of infection as part of a mechanism to mediate macrophage cytotoxicity against different infectious agents [41].
The increased CD69 expression, cell prolif-eration
and NO production were observed with the probiotic L. casei and the prebiotic agave fructans.
These results suggest that the agave fructans/L. casei mixture might have immunomodulatory effects on T lymphocytes, as monocytes/macrophages play a cen-tral role in immune regulation through the presentation of antigen to T lymphocytes [42,43].
Notably, a direct interaction between immune system cells
and fructans was demonstrated, as the highest expression of CD69 on lymphocytes of stimulated PBMC was observed only in the presence of agave fructans,
and the effect was even greater than that presented by PBMC stimulated with either LAB alone or fructans/LAB (Fig. 2a).
The activated cells interacted with other immune-component cells
and might induce different immune sig-nals and responses.

3.3. Transcription factor expression in lymphocyte differentiation of stimulated PBMC Previous reports indicate that prebiotics/probiotics induce a shift in the type of T helper immune response against some anti-gens [44,45].
Therefore, the relative expression of lymphocyte evaluated.

fructans (chicory inulin and agave fructans) showed the increased expression of Tbet
and FOXP3 transcription factors (Fig. 5b).
These results suggest that prebiotics directly or indirectly stimulate intestinal host defenses to produce a balanced T helper cell response
and prevent an imbalance that contributes to manifesta-tion of clinical disease [47]. However, additional studies are needed to show that prebiotics might directly induce the polarization of T helper responses.
Interestingly, an interaction between probiotic
and prebiotic stimuli was observed (Fig. 5c).
The differentiation induced through L. casei was greatly diminished, specifically for Tbet (Th1 response),when combined with agave fructans compared with L. casei alone.
In addition, B. lactis inducedGATA3 expression (Th2 response) when combined with agave fructans compared with B. lactis alone (Fig. 5a and c).
Notably, this phenomenon was not induced using inulin mixtures (Fig. 5a and c), as this compound did not specifically induce Th1 or Th2 cells (Tbet and GATA3 respectively).

These results suggest that agave fructans might specifically modulate the shift of T helper cells, depending on the LAB strain used.
Thus, agave fruc-tans should be used as a dietary product, as this compound might stimulate the adaptive immune response in a Th1 or Th2 direc-tion.
The induction of Th1 responses might prevent Th2-related immune disorders, such as allergies [44,45]
and the induction of Th2 responses to prevent or treat Th1 immune disorders, such as inflammatory bowel disease (IBD). Moreover, the induction of Th1 cells might treat the age-related decline in lymphoid cell activity 4. Conclusions
This study is the first investigation of the in vitro effect of agave fructans on the immune system.
Agave fructans showed a prebi-otic effect comparable to that of inulin-type fructans, which have been widely distributed for foodstuffs
and are involved in the acti-vation
and selective differentiation of cells of the immune system through interactions with probiotics.
These results are promising for the agave fructans extracted from A. salmiana.
Further in vitro and in vivo studies are needed to complement these results
and enhance our understanding of the direct or indirect effect of these compounds on the immune system, resulting in the controlled
and proper application of fructans in the development of functional products.

1. บทนำ

2. วัสดุและวิธีการ
2.1 Fructans และจุลินทรีย์

และตัวทำละลายชนิดอินติลินชนิดซิออรีน (Sigma-Aldrich, St. Louis, MO, USA)
ถูกนำมาใช้และลักษณะเฉพาะของสารเหล่านี้แสดงไว้ในตารางที่ 1 [4,29,30] เชื้อแบคทีเรีย lac-tic (ไลบรารี) Loph (LAB) 2 สายพันธุ์ ได้แก่ Bifidobacterium lactis (SACCO BLC1, RAFF, Zapopan, Jal, Mexico)

3. ผลลัพธ์และการอภิปราย
3.1 การประเมินกิจกรรม prebiotic ของ fructans ลักษณะสำคัญของ prebiotics คือความสามารถในการเพิ่มการแพร่กระจายของจุลินทรีย์หรือโปรไบโอติกที่เป็นประโยชน์ใน microbiota colonic ของมนุษย์ [10] ในการศึกษาในครั้งนี้เราได้วิเคราะห์ฤทธิ์ทางชีวภาพของ A. salmiana fructans (Fructan A.s) ที่สกัดได้เมื่อเทียบกับการเตรียมสาร prebiotics แบบเดิม
